SEATTLE (AP) ? The expert skiers wasted no time after an avalanche hit and swept their friends down a steep slope in Washington state. They immediately turned on their emergency beacons and began searching for signs of life.

Powder Magazine senior editor John Stifter, who witnessed the slide that killed three of his skiing companions Sunday, said one person survived by bear-hugging a tree and holding on as the snow barreled over him. Another skier who was caught in the slide was saved when she deployed an air bag designed to keep her afloat.

“It’s an absolute horror story,” Stifter said Monday.

The chances of surviving an avalanche once it has you in its grips are slim, experts say.

“The snow doesn’t really care how experienced you are. It’s not keeping track of experience level,” said Mark Moore, an avalanche meteorologist and director of the Northwest Weather and Avalanche Center, which warned of extreme avalanche danger Tuesday. “Once you’re in an avalanche, it has you at its mercy.”

Stifter identified the victims as Jim Jack, a well-known head judge for the Freeskiing World Tour; Stevens Pass marketing director Chris Rudolph; and Johnny Brenan, a Leavenworth contractor.

The Freeskiing World Tour and Utah’s Snowbird Ski and Summer Resort had scheduled a gathering at the resort Monday afternoon to remember Jack, whom Stifter described as generous, smart and influential in the ski industry.

The skiers were equipped with safety devices and kept track of each other as they strayed beyond the boundaries of the popular Stevens Pass Ski resort, about 90 miles northeast of Seattle. But the precautions still didn’t save some from getting trapped, highlighting the risks of backcountry activity during a season of heightened avalanche dangers in the West.

Sunday’s avalanche was relatively large, Moore said. The Northwest Weather and Avalanche Center rate Sunday’s avalanche danger as considerable to high. Heavy snow had fallen in the Cascades on Saturday with widespread avalanches and strong winds, all red flags, Moore said.

“Most of our avalanches here are storm-related, so we get most of our avalanche activity during or immediately after a storm,” Moore said. “It’s very sad to have accidents like this happen. No matter how good the snow is, you still have to be objective about risk,” he added.

Statistics show that 93 percent of avalanche victims can be recovered alive if dug out within 15 minutes, but survival rates drop quickly as time passes, according to the Utah Avalanche Center. After 45 minutes, only 20 to 30 percent of victims are alive. After two hours, few survive. People die because their carbon dioxide builds up in the snow around their mouth and they quickly die from carbon dioxide poisoning.

The Tunnel Creek canyon ? where a snowboarder died in an avalanche last year ? is outside the boundaries of Stevens Pass ski resort, but the area can be accessed by taking one of the resort’s lifts to the top and hiking a short distance. The area is not controlled for avalanches.

Stifter said he and Jack skied in the resort until about 11:15 a.m. Sunday when they met up with Rudolph. Stifter was in Washington state on an assignment, but this “was just a fun run with friends.”

He said he read the avalanche report that morning and knew avalanche dangers were considerable. He and others talked about it and determined they could ski it safely, he said. Jack and Rudolph had both skied the area countless times, he said.

Three of the 15 in the initial group peeled off and went a different way. Twelve others headed down, pairing up and skiing one by one, leapfrogging each other.

Each carried shovels and avalanche probes and wore avalanche beacons, a standard rescue device that allows rescuers to locate the signal of a victim if buried.

Stifter watched as Jack made three turns, and then he saw a pocket of snow pop out. “Holy smokes! This is huge,” Stifter recalled, when they realized how huge the avalanche was.

“We immediately pulled out our transceivers, designated a leader and spaced out 30 feet, zig-zagging all the way down,” hoping to pick up their friends’ signals, Stifter said.

Eventually, they picked up on the first signal and began digging furiously. They found Rudolph face down. Stifter performed CPR for about 30 minutes to no avail. Another group worked on digging out Brenan and Jack.

Professional skier Elyse Saugstad told NBC’s “Today” show she’s convinced the air bag she deployed immediately ? which she carried in a backpack and deployed with a lever by her chest ? saved her life.

“It’s lifting you kind of up above the avalanche,” Saugstad said Monday. “It’s not like you’re taking an inner tube ride down some snowy field. … It feels like you’re in a washing machine.”

Only Saugstad had an air bag, Stifter said. Air bags range from about $600 to $1,000. They have been widely used in Europe with reports of high survival rates, but they have become popular in the U.S. only recently.

An avalanche beacon, shovel and probe are among the mandatory rescue items for those heading into the backcountry, but experts say it’s best to avoid avalanches entirely.

“The truth is, if you have to use your beacon, it means you’ve made a big mistake,” said Benj Wadsworth, executive director of the Friends of the Northwest Weather and Avalanche Center, a nonprofit that works with the center to promote avalanche education and safety. “The focus of avalanche education is to keep you out of avalanches in the first place.”

Adds Moore: “There are all of these technological things that will help us, but they’re not a talisman that you can wave at the snowpack. You can’t wave your beacon or your air bag at the snowpack. It’s not going to make you safe. It’s going to help you when get in trouble.”

Telomere failure, telomerase activation drive prostate cancer progression

ScienceDaily (Feb. 20, 2012) ? Genomic instability caused by an erosion of the protective caps on chromosomes, followed by activation of an enzyme that reinforces those caps, allows malignant cells to evade destruction and acquire more deadly characteristics, researchers report in an Online Now article at the journal Cell.

In a strain of mice engineered to develop prostate cancer, all mice that went through this two-step process developed lethal cancer and 25 percent had the disease spread to the spine. Two groups of mice that avoided this cycle developed only precancerous lesions or localized prostate cancer.

A comparative analysis of genetic changes in the metastatic mouse tumors and those found in metastatic human prostate cancer identified the Smad4 gene as a driver in bone metastasis. Fourteen other genes were found to be associated with human prostate cancer prognosis.

The research focused on telomeres — repeat nucleotide sequences at the tips of chromosomes that prevent genomic damage during cell division. Telomeres shorten with each cell division, eventually permitting genomic instability in the cells that normally causes these abnormal cells to die.

In cancer the enzyme telomerase becomes activated and lengthens telomeres, preserving damaged cells to survive and reproduce. Telomerase is inactive in normal cells.

Telomerase activation confers new strengths on tumors

“These in vivo mouse studies, together with human and mouse prostate cancer genomic data, provide evidence that telomere dysfunction plays a critical role in prostate cancer initiation and progression,” said co-senior author Lynda Chin, M.D., professor and chair of The University of Texas MD Anderson Cancer Center’s Department of Genomic Medicine.

“Our studies also show that telomerase activation after genomic instability caused by telomere dysfunction enables evolving cancers to progress and acquire new biological properties, including central features of advanced human prostate cancer,” Chin said.

Chin, co-senior author and MD Anderson President Ronald DePinho, M.D., and colleagues conducted this research while at Dana-Farber Cancer Institute in Boston.

Telomere dysfunction, fired-up telomerase, cause bone metastasis

The team took a strain of mice with both the p53 and pten tumor-suppressing genes knocked out that normally develop nonmetastatic prostate cancer and engineered some to express telomerase. They were cross-bred for several generations.

Control mice with intact telomeres (either wild-type mice or those with telomerase expressed) avoided the genomic instability caused by telomere shortening. All of these mice developed locally invasive, nonmetastatic prostate cancer.

Mice without telomerase were subject to telomere dysfunction and genetic changes and developed precancerous high-grade prostate intraepithelial neoplasia (HPIN) but 60 percent of them did not progress to prostate cancer. Signs of programmed cell death triggered by genetic abnormalities abounded in this group.

Mice subject to telomere dysfunction, genomic instability and telomerase activation also developed HPIN but then progressed to lethal bulky tumors, with 5 of 20 developing spinal metastases that were not seen in the genome-stable mice.

“Not only did telomerase reactivation bypass the cancer progression block that arises with telomere dysfunction, it also conferred a new property — bone metastasis — that was not seen in tumors that did not go through telomere dysfunction followed by telomerase reactivation,” said first author Zhihu Ding, Ph.D., formerly of Dana-Farber and now with Sanofi-Aventis, Inc.

“This provides the first genetic evidence that telomerase reactivation and genome stabilization are necessary to drive full malignant progression in epithelial cancers,” Ding said.

Aligning genetic alterations in mice and humans

Chin, Ding and colleagues analyzed gene copy number aberrations — genes deleted or amplified — in 18 advanced tumors from the mice and 194 human prostate tumors.

They found 22 of the 94 copy number alterations involving deletion or amplification of 741 genes identified in mice were similar to those found in humans. A series of analyses of changes found in bone metastases pointed to deletions of the Smad4 tumor-suppressor gene, which regulates the transforming growth factor beta (TGF-?) pathway.

The team took this finding back to the mouse model with tumor suppressors p53 and pten knocked out. These mice don’t develop bone metastasis, but when the researchers also knocked out Smad4, more aggressive tumors developed, including bone metastasis in three of 24 mice.

Genes prognostic for human prostate cancer

Next, they looked at 14 genes in nine molecular pathways found to be enriched in bone metastasis to see if they were prognostic for recurrence (as measured by PSA levels after surgery) among 140 prostate cancer patients.

The 14-gene set was significantly prognostic of biochemical recurrence, providing evidence of their biological relevance to human prostate cancer, but the researchers noted that their individual contributions and mechanisms of action will require further research.

“Overall, our findings validate the integrative approach of employing genotype-phenotype correlations found in the mouse model with the power of genomic and bioinformatic analyses to discover and explain molecular mechanisms that drive prostate cancer,” said co-first author Chang-Jiun Wu, Ph.D., a postdoctoral fellow in MD Anderson’s Department of Genomic Medicine.

A two-and-a-half-year-long suspected archaeological fraud involving thousands of forged Greek and Etruscan artefacts, a hospital x-ray machine, a philanthropic aristocrat and a sophisticated network of forgers has come to an abrupt end after police raids late last year on two homes belonging to alleged members of a gang. Seven arrests were made and a further seven suspects are under investigation.

The gang was allegedly led by Edoardo David, a renowned archaeologist who often worked as a consultant for the archeological division of the Soprintendenza for the Lazio region (the local arm of Italy?s ministry of culture), and his two principal associates, Massimo Monaco, a former boxer, and Mariano Capomaggi, all of whom are being held at the Regina Coeli prison in Rome awaiting trial.

Four other people are under house arrest: Enrico Corradi, an antiques dealer from Aprilia, near Rome; Massimo Bordo, a ceramics expert, and Massimiliano Congiu, both from Tarquinia, near Rome; and Enrico Diomedi, a nurse who allegedly helped the gang produce thousands of suspected fakes by giving them access to an x-ray machine in the hospital at Tivoli, also near Rome.

The x-rays interfered with the outcome of any thermoluminescence testing of these modern pieces. Thermoluminescence is the faint light emitted by a ceramic that has been fired at a sufficiently high temperature. This light can be measured to establish the age of the piece; the brighter the light, the older the piece. Test results are invalidated if the piece is exposed to radiation.

Between March 2009 and April 2010, Marquis Roberto Bilotti Ruggi d?Aragona, an aristocratic philanthropist and antiques collector from Rome, who hoped to expand his collection and establish his own family museum of antiquities, spent ?600,000 on around 300 works that the police believe to be fakes.

He also says that he exchanged many of his own valuable items, including family heirlooms and antique furniture, with the gang for a number of artefacts he believed to be real. The gang is believed to have acquired more than ?1m worth of items from his collection. The marquis eventually became suspicious about the sheer number of rare artefacts he was being offered. He finally contacted the police in April 2010 after he was assaulted and robbed by unidentified assailants who broke into his home to steal a historic vase depicting the robing of Achilles, an artefact he had repeatedly refused to trade with the alleged forgers.

The police raided two properties belonging to Edoardo David and Massimo Bordo. They found more than 2,000 authentic artefacts at David?s property. The provenances of all the items have yet to be fully investigated, although some were purchased from the marquis, and the police have identified many others as stolen. The police also found 3,000 allegedly forged artefacts ready for market at Bordo?s property.

Bordo, an expert in ceramics, allegedly used complex techniques to produce the fakes, grinding up a small amount of authentic antique ceramic material and blending it with the ordinary, modern mix from which he would then shape the artefacts.
